Bioindicators of pollutant exposure in the northwestern Mediterranean sea ArchiMer

Several bioindicators were used to evaluate the biological and genotoxic effects of marine pollutants near large coastal cities in the northwestern part of the Mediterranean Sea. Three target species of teleosts were selected: red mullet Mullus barbatus and 2 types of comber (Serranus hepatus and S. cabrilla). Induction of ethoxyresorufin-O-deethylase (EROD) activity specific for polycyclic aromatic hydrocarbons (PAH) and polychlorinated biphenyls (PCB) was measured in the livers of the fish, and inhibition of acetylcholinesterase (AChE) by organophosphorus insecticides and carbamates was measured in their muscle tissues. There are few studies of ochratoxin A (OTA) genotoxicity in experimental animals and the results obtained with cell cultures are inconsistent, although the carcinogenic potential of OTA for the kidney of experimental animals has been well established. DNA damage in dab (Limanda limanda) and haddock (Melanogrammus aeglefinus) from European seas ArchiMer

Dab (Limanda limanda) and haddock (Melanogrammus aeglefinus) were collected from coastal and offshore locations in the Baltic (dab only), North Sea (haddock from one location only) and Iceland. Blood was analysed for DNA strand breaks using the comet assay and liver samples for DNA adduct concentrations. DNA strand breaks were at background levels in dab from the two Iceland locations and from the Dogger Bank. The highest levels were observed in dab from the Firth of Forth, Ekofisk and the German Bight. Hepatic DNA adducts in dab were highest at Ekofisk, in the Baltic and Dogger Bank, below detection limit in dab from Iceland and low in dab from the Firth of Forth and German Bight. Genotoxicity and activation of cellular defenses in transplanted zebra mussels Dreissena polymorpha along the Seine river ArchiMer

The aim of the present study was to confirm the relevance of studying DNA adduct formation in a field study. In that context, freshwater mussels Dreissena polymorpha, collected in a reference station, were transplanted in different sites with a pollution gradient. After one and two months, mussels were collected and DNA adduct formation was analyzed using the 32P post labelling technique on both gills and digestive glands. In addition, the expression of genes involved in the detoxification system (catalase (CAT), superoxide dismutase (SOD), glutathione S-transferase (GST), HSP70, aryl hydrocarbon receptor (AHR), P glycoprotein (PgP), metallothionein (MT)) was assessed by RT-PCR. Histopathological lesions and DNA adducts in the liver of European flounder (Platichthys flesus) collected in the Seine estuary versus two reference estuarine systems on the French Atlantic coast ArchiMer

An epidemiological survey was conducted in the Seine estuary and in two smaller and relatively preserved estuaries on the French Atlantic coast in order to estimate the occurrence of liver lesions in European flounder, Platichthys flesus, and also to seek putative risk factors for the recorded pathologies. Four hundred and seventy-eight fish of both sexes and of different size ranges were sampled in the three studied areas, 338 of which in the Seine estuary. All fish were examined for histopathological liver lesions, while DNA adducts and otoliths were analyzed on a subsample. Induction and elimination of bulky benzo[a]pyrene-related DNA adducts and 8-oxodGuo in mussels Mytilus galloprovincialis exposed in vivo to B[a]P-contaminated feed ArchiMer

With the aim of developing biomarkers of polycyclic aromatic hydrocarbon (PAH) genotoxicity for biomonitoring of the marine environment, the formation of DNA adducts and oxidative DNA damage was studied in the sentinel organism Mytilus. Mussels M. galloprovincialis were exposed for 21 d to different doses of benzo[a]pyrene (B[a]P) (50 and 100 mg B[a]P kg(-1) dry wt mussel d(-1)) via trophic intake. The kinetics of B[a]P concentration in whole mussel tissues was determined in both groups, and a dose-response study was conducted for both gill B[a]P-related DNA adducts and 8-oxo-7, 8-dihydro-2'-deoxyguanosine (8-oxodGuo) formation in gill and digestive gland DNA. Three biomarkers [ethoxyresorufm-o-deethylase (EROD), DNA adducts and micronuclei (MN) were studied for biomonitoring applications along French coasts. EROD was developed at an experirnental stage in two pilot sites, Seine Bay and Fos sur Mer, using the dab (Limanda limanda), dragon et (Callionymus lyra), red mullet (Mullus barbatus) and comber (Serranus hepatus) as target species. Higbest EROD activity was observed in the subestuarial part of Seine Bay and at the stations closest to sources of pollution from Cortiou and Fos sur Mer. The benzo[a]pyrene is a polycyclic aromatic hydrocarbon known to be genotoxic, mutagenic and carcinogenic in higher vertebrates. The aim of this study was to evaluate in vitro the enzymatic and genotoxic effects of BaP in a benthic fish species, Solea solea. Sole hepatocytes were exposed to BaP in order to measure the modulation of ethoxyresorufin-o-deethylase (EROD) activity and the DNA strand breaks induced by BaP metabolism. Exposures were performed in both culture flasks and microplate wells in order to check for the possible miniaturization of the exposure system.
